Genome Mining and Screening of Anaerobic Bacteria for Sulfur-containing Natural Products

Natural products are an important source for drug development and in recent years an increasing number of novel scaffolds have been discovered from anaerobic bacteria. Indeed closthioamide (CTA), produced by the anaerobic bacterium Ruminiclostridium cellulolyticum is a rare example of a thioamidated natural product and is a potent antibiotic. The recent elucidation of CTA biosynthesis identified the ctaC gene, which encodes the enzyme that catalyzes the amide thionation. Subsequent bioinformatic analysis identified multiple other anaerobic bacteria with ctaC homologs. These strains will be screened for sulfur-containing natural products with UHPLC-HRMS.
